I have many computers/servers. How would I create a central update location so that all updates are downloaded to that location and all workstations/servers get their updates from there as opposed to the Internet?

I think there are some scripts on these ClamWin forums that you could use. Try some searching in the various sections. A search on the web for "ClamWin update scripts" or similar might also be a good idea.
ClamWin is only a on-demand scanner, and the Clam AV signature database includes detections for only a small subset of malware. The ClamWin developers recomment that you use a real-time primary AV and employ ClamWin as a "backup" scanner.
