There can be several reasons why the signature database can't be initialized. One common reason is that the database has become corrupted for some reason. To fix this, delete every file in the ClamWin database in the Programsx86\data\ClamWin folder and then do a manual update of the signatures. If that doesn't work, try an uninstall and then a re-install of ClamWin (run it as an administrator). Let us know how it goes.
